ATHENS, Greece (AP) ? Greece?s Finance Ministry says the country?s total central government debt stood at ?303.5 billion ($372.67 billion) at the end of July 2012, up from ?280.2 billion at the end of the first three months of this year.

The country has been struggling with a severe financial crisis since late 2009, and is dependent on international rescue loans from the International Monetary Fund and other European countries that use the euro. In return for billions of euros in loans, Greece has imposed stringent austerity measures in an effort to make its debt sustainable and allow it to return to borrowing on the market, from which it has been barred by sky-high interest rates.

The finance ministry released the figures Friday, which also showed that government debt hit its highest level ? ?367.9 billion ? in the fourth quarter of 2011.

Your Special Challenges

Lifting and carrying: Your doctor may have advised you to take it easy with the heavy stuff. It's perfect excuse to delegate and supervise. Have someone else set up the tent and or carry the heaviest pack.

The Business of Peeing: It's not so easy for women as it is for men. First we have to find a private spot and then peel off half of our clothes. Getting up and down out of a squat might be a challenge. Some moms prefer camping at a spot with restrooms. Others make building an outhouse a high priority. The rest just deal with it or find a smooth fallen log to help out.

Hygiene: You might not be able to shower or find a clean lake for a swim. During pregnancy, women tend to sweat more. Bring along supplies for a refreshing sponge bath.

Safe Water: Bring you own water or drink from a safe water source. Intestinal problems from water borne illness can be serious for both mom and baby. The iodine tablets used to treat water aren't considered safe during pregnancy. If you use local water, boil it the recommended time for your camp's elevation.

Metabolic protein wields phosphate group to activate cancer-promoting genes

ScienceDaily (Aug. 16, 2012) ? A metabolic protein that nourishes cancer cells also activates tumor-promoting genes by loosening part of the packaging that entwines DNA to make up chromosomes, a team led by scientists at The University of Texas MD Anderson Cancer Center reports in the Aug. 16 issue of Cell.

Working in cell lines and mouse models of glioblastoma multiforme, the most lethal form of brain tumor, senior author Zhimin Lu, Ph.D., associate professor of Neuro-Oncology at MD Anderson, and colleagues show that pyruvate kinase M2 (PKM2) fuels tumor growth by influencing a histone protein.

DNA is packaged in and spooled around histone proteins. The researchers found that PKM2 tags histone H3 with a phosphate group (one atom of phosphorus, four of oxygen) in a specific location called T11.

'No phosphorylation of H3, no tumor'

This phosphorylation leads to activation of the tumor-promoting genes, increased tumor cell reproduction and formation of tumors, Lu said. "If there's no phosphorylation of H3, there's no tumor. It's that crucial to glioblastoma formation."

An analysis of 85 human glioblastomas indicated that higher levels of PKM2 expression in the cell nucleus and of H3 phosphorylation are correlated with shorter survival. A separate analysis showed higher levels of H3 phosphorylation associated with higher grade tumors in a comparison of 30 low-grade tumor samples and 45 high grade glioblastomas.

"Histone 3-T11 phosphorylation has great potential to serve as both a prognostic marker and a guide for the use of PKM2-inhibiting therapies once they are developed," Lu said.

PKM2 has long been known for its well-established role in aerobic glycolysis -- the processing of glucose into energy that solid tumors, glioblastomas in particular, rely on heavily to survive and grow. Lu and colleagues have been teasing out the mechanisms of PKM2's other role -- the transcription and activation of genes.

It all starts with EGFR

When the epidermal growth factor receptor (EGFR) on the cell's membrane is activated by a growth factor, the PKM2 protein moves into the cell nucleus, where it binds to the promoter regions of genes. Other proteins called transcription factors attach to a gene's promoter region to activate it.

Cancer cells have high levels of EGFR on the cell surface, relaying growth signals from outside the cell inside. EGFR is itself a target of some cancer drugs.

A series of experiments by the research team uncovered the following molecular steps:

* After EGFR activation, PKM2 binds to histone H3 and attaches a phosphate group at T11.

* This separates another protein called histone deacetylase 3 (HDAC3) from the promoter regions of the genes CCND1 and MYC. HDACs block gene activation.

* With the HDACs gone, histone H3 acquires an acetyl group, which facilitates gene activation.

"This series of events only occurs when H3 is phosphorylated by PKM2," Lu said.

Blocking phosphorylation prevents brain tumors in mice

CCND1 expresses the protein cyclin D1, a cell cycle regulator. The MYC gene is frequently mutated in cancer, leading to overexpression of the transcription factor Myc, which in turn causes unregulated expression of many other genes.

Mouse experiments of EGFR-driven glioblastoma using reconstituted H3 histones, one normal and one with a mutant version of H3-T11A to prevent phosphorylation by PKM2, confirmed the relationship. Mice injected with normal, or wild type, H3, had an average tumor volume of nearly 40 cubic millimeters, while those with disabled T11A, blocking the phosphorylation point for PKM2, had no tumors.

"Our findings establish PKM2 as a histone kinase, which directly regulates gene transcription and controls cell cycle progression and proliferation of tumor cells" Lu said. Kinases are a class of proteins that attach phosphate groups to other proteins.

Alfred Yung, M.D., all of MD Anderson's Department of Neuro-Oncology; David Hawke, Ph.D., of MD Anderson's Department of Molecular Pathology, Kenneth Aldape, M.D., of MD Anderson's Department of Pathology; Dongming Xing of School of Life Sciences, Tsinghua University in Beijing; and Tony Hunter, Ph.D., of the Salk Institute for Biological Studies in La Jolla, Calif.

Kim Kardashian and Kris Humphries may never get divorced.

The former couple continued to battle in court yesterday, with all signs pointing to Humphries' legal team dragging out these proceedings for as long as possible in an attempt to prove Kim somehow defrauded him into marrying her... or to simply irritate the heck out of his estranged wife.

Kardashian, meanwhile, is allegedly telling friends she just wants her divorce finalized - NOT because she wants to marry Kanye West, but because Kris is a "cancer" she just wants to be rid of.

"Kris is the first person I ever had to break-up with and that f-cked with my emotions," Kim is supposedly telling friends, according to TMZ, going into detail about the pair's experience filming Keeping Up with the Kardashians together:

"He told our producers he would destroy my career and me if the show wasn't edited right."

But that strategy will "backfire" on Humphries, Kardashian says, if he keeps claiming she was to blame for their marriage falling apart.

She cites unaired footage from the E! series that is still out there and which depicts Kris as a "manipulative, vindictive, petty, fame-hungry jerk."

And, we can only presume Kim added, no way I'll let him beat me at my own game!

Mayim Bialik Severely Injured in Car Accident, May Lose a Finger

Actress Mayim Bialik was involved in a car accident this afternoon in Los Angeles, sustaining a severe injury that may result in the loss of a finger.

Sources tell TMZ that the crash took place around noon at the corner of Hollywood Blvd. and La Brea. The Big Bang Theory star was reportedly driving alone in a white Volva and was struck by another vehicle, which was carrying tourists from Chile.

An ambulance soon arrived and witnesses say there was "tons and tons of blood everywhere," adding that Bialik's finger was "almost completely severed… it was just hanging there."

The 36-year old is being treated at the hospital while we type. We send her our very best wishes.

UPDATE: Bialik has Tweeted that she is in a lot of pain, but will not lose any digits.

Scent of sex and death attracts virgin female beetles

For one insect, the allure of sex pheromones is not enough to attract a mate. Throw in the scent of rotting flesh, however, and the cocktail becomes irresistible.

We already know that newly hatched male hide beetles (Dermestes maculatus) are attracted by benzyl butyrate, a chemical that is released in relatively high concentrations by cadavers in the late stage of decay. Their presence is such a predictable part of the decaying process that forensic scientists use them to estimate time of death.

Less clear, though, is what attracts recently hatched, virgin females. To find out, Christian von Hoermann, a forensic entomologist at Ulm University, Germany, and his colleagues exposed virgin females to several potentially alluring scents. They found that neither the male sex pheromones nor the scent of a pig's cadaver at various stages of decay were significantly more attractive to the virgin females than the scent of a control solvent. But a combination of dead pig scent and male pheromone had the females flocking.

The behaviour makes sense: a rotting animal is the ideal place for healthy beetle larvae, and what better place to mate than the place where your kids will grow healthy and strong?

"This scent of love is an honest bouquet for the appropriate decomposition stage for larval development," says von Hoermann.

Tristram Wyatt at the University of Oxford says that similar behaviour is seen in other insects that depend on patchy resources. Once they find a suitable source of food, they release pheromones to attract suitable females, who are happy to get both food and a male at the same place. "For example, males of the scorpion fly Panorpa pheromone-call when they have an insect food item gift, while male papaya fruit flies call from fruits."
